PNC-27 vs SS-31

PNC-27 and SS-31 are studied in overlapping research areas, which is why they are frequently compared. This is a neutral side-by-side reference drawn from published preclinical literature and laboratory handling data.

Working windowShort — prepare working solutions close to use.Commonly worked with for 3-4 weeks at 2-8 °C.
Lead degradation routeAggregation of the amphipathic chain at the air-liquid interface or high concentration.Adsorptive loss to plasticware from the cationic charge — the main practical concern.

How they actually differ

Comparing the two: PNC-27 is membranolytic fusion peptide (32 residues): p53-derived domain + membrane-penetrating leader, while SS-31 is mitochondria-targeting aromatic-cationic tetrapeptide (cardiolipin-binding) — different molecular classes with different handling consequences; they call for different primary diluents (sterile water for a stock, then dilution into assay buffer versus bacteriostatic water (0.9% benzyl alcohol)); their leading degradation routes differ (aggregation of the amphipathic chain at the air-liquid interface or high concentration. for PNC-27, adsorptive loss to plasticware from the cationic charge for SS-31), so the storage precautions that matter are not the same; their practical working windows differ once reconstituted. The sections below set out each in full.

PNC-27 — origin

PNC-27 is a designed 32-residue peptide fusing a p53-derived region that binds HDM-2 with a membrane-penetrating leader sequence. It is studied strictly in vitro for a reported ability to form pores in the membranes of cancer cells that display HDM-2 at their surface, while reportedly sparing normal cells.

SS-31 — origin

SS-31 (Elamipretide) is a Szeto-Schiller aromatic-cationic tetrapeptide (D-Arg-2',6'-dimethyltyrosine-Lys-Phe-NH2) that concentrates in the inner mitochondrial membrane and binds cardiolipin, the lipid unique to that membrane. Its structure is the reason it is studied specifically at the mitochondrion rather than as a general antioxidant.

PNC-27 research themes

HDM-2 cell-surface targeting

Studied for binding HDM-2 displayed on the surface of certain cancer cells in vitro.

Membrane pore formation

Examined for forming transmembrane pores that lead to lysis of targeted cells in culture.

Selectivity in vitro

Investigated for a reported preference for tumour cells over normal cells in preclinical cell-culture models.

SS-31 research themes

Cardiolipin binding

Studied for selective association with cardiolipin in the inner mitochondrial membrane, the basis of its mitochondrial targeting.

Electron transport efficiency

Examined for effects on mitochondrial energy production in research models of mitochondrial dysfunction.

Oxidative stress

Investigated around reactive-oxygen-species handling at the mitochondrion.

PNC-27 handling

  • For laboratory in-vitro use only — this compound is studied strictly in cell-culture research.
  • Prepare a stock and dilute into the assay system rather than dissolving at high concentration.
  • Aliquot and refrigerate; prepare fresh working dilutions.

SS-31 handling

  • Reach room temperature before opening.
  • Use low-bind labware and buffers at low working concentrations to limit adsorption of the cationic peptide.
  • Aliquot and refrigerate.
