How to Read a FOXO4-DRI Certificate of Analysis (COA)

A Certificate of Analysis is only useful if you know which number on it matters for the compound in front of you. For FOXO4-DRI, the informative checks are not the same as for a generic short peptide.

Overview

Look at two numbers on the certificate: HPLC purity and how many milligrams are actually in the vial. Our batch FOX4-401-002 tested at 99.724% purity and 12.37 mg (Janoshik #93523). If a mass is reported, it should be about 5358.

What FOXO4-DRI actually is

FOXO4-DRI is a lab-made peptide that blocks two proteins, FOXO4 and p53, from holding onto each other inside worn-out "senescent" cells. In the 2017 study that introduced it, breaking that grip caused senescent cells in the dish to self-destruct while healthy cells were left alone. It is studied in cell and animal research on how senescent cells build up with age and damage. You will also see it spelled FOX04-DRI (with a zero) or sold as Proxofim — same molecule.

What to check on the COA

Check the HPLC purity and the measured content per vial. For a 46-residue, highly cationic peptide the gross powder weight includes counter-ion (typically trifluoroacetate) and water, so measured peptide content is the more informative number. Where mass data is given, the target is about 5358 Da.

Characteristic impurity profile

Deletion and truncated sequences from a long solid-phase synthesis; deamidated forms eluting close to the main peak.

What FOXO4-DRI is studied for

FOXO4–p53 interaction in senescent cells

The founding study showed that senescent cells rely on FOXO4 holding p53 in the nucleus. Interfering with that interaction caused p53 nuclear exclusion and intrinsic apoptosis selectively in senescent cells in culture (Baar et al., Cell 2017).

Chemotoxicity and ageing mouse models

The same study examined doxorubicin-induced senescence, fast-ageing XpdTTD/TTD mice and naturally aged mice, reporting changes in fur density, fitness and renal markers.

Senescent Leydig cells

Studied in hydrogen-peroxide-induced senescent TM3 Leydig cells and aged mice for effects on the testicular microenvironment (Zhang et al., Aging 2020).

Expanded human chondrocytes

At 25 µM in vitro it removed a large share of late-passage (senescent-enriched) human chondrocytes while sparing early-passage cells, without improving cartilage-forming capacity (Huang et al., Front Bioeng Biotechnol 2021).
