What FOXO4-DRI actually is
FOXO4-DRI is a lab-made peptide that blocks two proteins, FOXO4 and p53, from holding onto each other inside worn-out "senescent" cells. In the 2017 study that introduced it, breaking that grip caused senescent cells in the dish to self-destruct while healthy cells were left alone. It is studied in cell and animal research on how senescent cells build up with age and damage. You will also see it spelled FOX04-DRI (with a zero) or sold as Proxofim — same molecule.

Why these conditions, specifically
Because every residue is a D-amino acid, ordinary proteases barely cleave it — chemical routes (deamidation, oxidation) and surface loss matter far more than enzymatic breakdown.
The main route to be aware of: deamidation at the Asn-Gly motif in the sequence — the fastest-deamidating dipeptide in peptide chemistry.

Aliquot on first reconstitution and freeze aliquots once; avoid repeated freeze–thaw cycles, which concentrate the peptide and salts at the ice interface.
What FOXO4-DRI is studied for
The founding study showed that senescent cells rely on FOXO4 holding p53 in the nucleus. Interfering with that interaction caused p53 nuclear exclusion and intrinsic apoptosis selectively in senescent cells in culture (Baar et al., Cell 2017).
The same study examined doxorubicin-induced senescence, fast-ageing XpdTTD/TTD mice and naturally aged mice, reporting changes in fur density, fitness and renal markers.
Studied in hydrogen-peroxide-induced senescent TM3 Leydig cells and aged mice for effects on the testicular microenvironment (Zhang et al., Aging 2020).
At 25 µM in vitro it removed a large share of late-passage (senescent-enriched) human chondrocytes while sparing early-passage cells, without improving cartilage-forming capacity (Huang et al., Front Bioeng Biotechnol 2021).
