Choosing a diluent for Cagrilintide

Reported aqueous solubility varies widely between reagent suppliers — from sparingly soluble in PBS at pH 7.2 to freely soluble in water with sonication — so treat any single published figure as buffer-, pH- and lot-dependent rather than a fixed property. The behaviour is characteristic of lipidated peptides: the C20 diacid that drives albumin binding also drives self-association, which makes apparent solubility sensitive to conditions. Introduce diluent slowly and allow time before concluding a vial has failed to dissolve.

Handling notes for Cagrilintide

  • Never shake. Swirl gently and allow the cake to dissolve in its own time.
  • Do not freeze reconstituted solution.
  • Add diluent down the vial wall rather than directly onto the cake.
  • Treat visible particulate or a settling precipitate as a rejected vial — for an amylin analogue this is the observable end of the fibrillation pathway, not a cosmetic issue.

Freeze–thawAvoid freezing reconstituted material. For an amylin analogue the freeze–thaw concern is not only interfacial aggregation but nucleation of the fibril pathway the molecule was engineered to resist — and fibril formation, once seeded, does not reverse on rewarming.
