What to Check When Your Cagrilintide Arrives

Not every irregularity on arrival is a defect, and not every compound is defective under the same conditions. Here is what actually matters when a Cagrilintide shipment arrives.

Lipidated 37-residue amylin analogue, Cys2–Cys7 disulfide, C-terminal prolinamideMetabolic

Overview

The dry powder travels well and a warm journey is usually fine. What is not fine is a vial that arrives with the contents already melted, collapsed or partly liquid — that means moisture got in, and it should be rejected rather than mixed.

What Cagrilintide actually is

Cagrilintide is an engineered copy of amylin — a hormone released from the pancreas at the same time as insulin. The natural version has a well-known problem: it clumps into fibres very readily, which is why nobody could make a long-lasting version of it for years. Cagrilintide is that problem solved, with six changes to the sequence and a fatty chain attached to make it last about a week instead of half an hour.

Supplied for laboratory research use only — not for human or animal use.

Research-grade Cagrilintide

Third-party tested by HPLC and LC-MS, ≥99% purity, with a Certificate of Analysis on every order. Ships across Canada.

Technical detail below

Arrival inspection for Cagrilintide

Inspect for cake integrity. The dry material is robust enough that ambient transit is generally acceptable, but any vial arriving with the cake liquefied, collapsed or partially dissolved has seen moisture and should be rejected rather than reconstituted.

Storage on arrival

On receiptSealed at -20 °C, protected from moisture. Supplier data sheets for the reagent-grade material specify sealed storage under nitrogen, away from moisture and light.
Once reconstitutedRefrigerate at 2–8 °C. As with other fatty-acid-acylated peptides, the acylation that extends half-life also contributes real solution stability.
Working windowCommonly worked with for 3–4 weeks at 2–8 °C.

Documentation to check

Confirm the mass near 4409 Da. Because the sequence carries a Cys2–Cys7 disulfide, correct disulfide pairing is a meaningful identity question that a purity percentage alone does not answer. As with other acylated peptides, aggregate content by size-exclusion chromatography adds information RP-HPLC cannot, since the organic mobile phase tends to dissociate soluble aggregates before they are detected.

What Cagrilintide is studied for

Amylin receptor pharmacology

Cagrilintide binds the calcitonin receptor and all three amylin receptors (AMY1, AMY2, AMY3), which is why the literature sometimes classes it as a dual amylin and calcitonin receptor agonist.

Amyloid-resistant peptide design

A worked example of engineering a fibril-prone hormone into a stable long-acting analogue — salt-bridge helix stabilisation plus beta-sheet-breaking prolines.

Protraction by acylation

The C20 diacid extends the half-life from minutes, for the short-acting analogue pramlintide, to roughly a week.

Combination with incretin agonists

The most-studied context: co-administration with semaglutide, investigated in the REDEFINE trial programme.
