DSIP — compound background

Nonapeptide (9 residues), strongly acidic

DSIP was isolated in the 1970s from the cerebral venous blood of rabbits in slow-wave sleep, in one of the more unusual isolation efforts in neuropeptide research. The name records the assay it was found by rather than a settled mechanism — its physiological role remains debated in the literature.

Solubility & reconstitution

Water-soluble, but distinctly more acidic than the other short peptides here: two acidic residues (Asp, Glu) and no basic residues give it a low isoelectric point. Solubility is therefore good at neutral and alkaline pH and drops as the solution approaches the pI in the acidic range — the opposite pH behaviour to cationic peptides like Selank.

Storage

Sealed at -20 °C. Keep dry; the acidic lyophilizate is hygroscopic. Refrigerate at 2–8 °C, protected from light on account of the N-terminal tryptophan.

Commonly worked with for 2–3 weeks at 2–8 °C.

Stability

DSIP combines two independent liabilities — a photo-labile N-terminal residue and an Asp-Ala isomerisation site. It rewards careful storage more than most short peptides.

Reading the Certificate of Analysis

Confirm the molecular ion near 848.9 Da. Watch for isomerised species, which are isobaric with the parent (identical mass) and therefore invisible to mass spectrometry — they show only as additional or broadened RP-HPLC peaks. This is a case where the chromatogram is more informative than the mass spectrum.
