IGF-1 Reconstitution Calculator

For in vitro research use only · Not medical advice

Preset to a 1 mg IGF-1 vial in 1 mL of diluent. A 1 mg vial in 1 mL yields 1 mg/mL. Dissolve first in dilute acetic acid, then dilute; aliquot for single use.

A 1 mg IGF-1 vial reconstituted with 1 mL gives 1 mg/mL. Measuring 0.100 mL gives 100 mcg per portion — about 10 portions from the vial.

Choosing a diluent for IGF-1

As a folded protein, IGF-1 dissolves poorly at neutral pH and is conventionally taken up first in a mildly acidic solvent, then diluted into the working buffer. A carrier protein is commonly used to limit adsorptive loss. Direct dissolution in plain water or PBS can leave incomplete dissolution — expected chemistry, not a defective vial.

PrimaryDilute acetic acid for initial dissolution
Alternative 1Then dilute into sterile water or PBS with a carrier protein
Alternative 2Bacteriostatic water for subsequent working dilutions

Handling notes for IGF-1

  • Dissolve first in dilute acetic acid, then dilute into the working buffer — never expect plain water to take it up fully.
  • Use a carrier protein and low-bind labware to prevent adsorptive loss.
  • Aliquot for single use; do not re-freeze reconstituted protein.

After reconstitution

StorageRefrigerate at 2-8 °C and use within a short window; single-use aliquots are strongly preferred because denaturation is irreversible.
Working windowShort — days rather than weeks — and shorter still with repeated handling.
Freeze–thawPoorly tolerant. Repeated freeze-thaw denatures the folded structure permanently, so aliquot for single use rather than relying on any freeze tolerance.
